The Role of Glycolysis in the Growth of Tumor Cells. Iv. the Basis of Glucose Toxicity in Oxamate-treated, Cultured Cells.

چکیده
During the course of experiments designed to establish the site of action of oxamate on HeLa cell growth (I), we found that the oxamate inhibition of growth was greatly intensified by the presence of glucose. This has now been observed in two different types of experiments. Under conditions during which growth is not dependent on added hexose (l), the growth is much more severely inhibited by oxamate when glucose is present. Where a hexose is required for growth, oxamate inhibits growth strongly when glucose is used but not at all when galactose is used. In this paper, we will try to elucidate the mechanism of this glucose toxicity.

Effective inhibition of nasopharyngeal carcinoma in vitro and in vivo by targeting glycolysis with oxamate.
Elevated aerobic glycolysis in cancer cells (Warburg effect) has been observed in many tumor types including nasopharyngeal carcinoma (NPC), which can often be detected clinically using FDG-PET. However, the role of glycolysis in supporting the growth of NPC cells and its therapeutic implications still remain to be investigated. In the present study, we showed that the LDH inhibitor oxamate sig...

Fibroblasts could be considered as connective tissue cells that are morphologically heterogeneous with diverse functions depending on their location and activity. These cells play critical role in health and disease such as cancer and wound by Production of collagen, fibronectin, cytokines and growth factors. Absence of insulin and other growth factors in serum deprivation condition and similar...
متن کاملThe Role of Glycolysis in the Growth of Tumor Cells
Otto Warburg demonstrated the high rate of aerobic glycolysisr of tumors in titro and postulated this as a unique requirement for neoplastic growth. (For a recent review see Aisenberg (I).) There is evidence that this metabolic anomaly is not due to a fundamental difference between normal and tumor cells but merely reflects their particular growth rates. For example, some years ago O’Connor (2,...
متن کاملCisplatin cytotoxicity is dependent on mitochondrial respiration in Saccharomyces cerevisiae
Objective(s): To understand the role of mitochondrial respiration in cisplatin sensitivity, we have employed wild-type and mitochondrial DNA depleted Rho0 yeast cells. Materials and Methods: Wild type and Rho0 yeast cultured in fermentable and non-fermentable sugar containing media, were studied for their sensitivity against cisplatin by monitoring growth curves, oxygen consumption, pH changes ...
